  • Sandwich immunoassay principle, where a target protein is immobilized between a capture antibody locked on magnetic microspheres and a detection antibody conjugated with a fluorophore.

RayPlexĀ® Line

RayBiotech’s RayPlexĀ® line is a microsphere-based (bead-based) multiplex assay platform designed for the simultaneous detection and quantification of numerous protein analytes in a single sample. It uses flow cytometry technology, combining the specificity of sandwich assays with the high throughput of multiplex systems.

Main Features

  • Multiplex Analysis: Allows simultaneous measurement of up to 25 or more proteins (such as cytokines, chemokines and growth factors) from a minimal sample volume (only 25 µl).
  • Universal Compatibility: Requires no dedicated or proprietary instrumentation; kits are compatible with most standard cytofluorometers equipped with blue (488 nm) and red (633/640 nm) lasers.
  • Efficiency: The operating protocol is rapid (about 4 hours total time) and significantly cheaper (5 to 10 times) than performing individual ELISA tests for each target.
  • Versatility of Targets: Pre-validated panels are available for human, mouse, rat and pig species, as well as the ability to create custom panels by choosing from more than 200 validated antibody pairs.

How It Works

The system is based on populations of microspheres of different sizes or levels of internal fluorescence. Each population is conjugated with a capture antibody specific for a given analyte:

  1. Capture: The beads are incubated with the sample (serum, plasma or cell supernatant).
  2. Detection: A cocktail of biotinalate detection antibodies is added, followed by streptavidin-phycoerythrin (PE).
  3. Readout: The cytofluorometer identifies the “bead” (and thus the analyte) based on its intrinsic fluorescent signature and quantifies its concentration based on the PE signal intensity.

Abcam ‘s Lightning-LinkĀ® conjugation kits represent one of the most innovative and rapid solutions for direct labeling of antibodies, proteins or peptides. This technology eliminates the need for complex post-conjugation purification steps, ensuring total (100 percent) recovery of valuable material.

Abcam’s Lightning-LinkĀ® kits are particularly popular in cytofluorimetry (FACS) because they allow researchers to create custom panels quickly, overcoming the limitations of commercial availability of directly conjugated antibodies.

Specific Benefits for Cytofluorimetry

  • Multiplex Panel Expansion: Allow the use of specific antibodies (rare or homemade clones) that are not commercially available already conjugated to the desired fluorophore.
  • Reduced Background: Direct conjugation eliminates the need for secondary antibodies, drastically reducing nonspecific binding and simplifying the design of the experiment (no need to worry about antibody source species).
  • Maximum Sample Recovery: Because there are no washing or purification steps, even very small amounts of antibody (up to 10 µg) can be marked, which is ideal for rare or expensive reagents.
  • Wide Choice of Fluorophores: The line includes all major dyes used in modern cytofluorometers:
    • Fluorescent proteins: APC, PE, PerCP (ideal for low-expression targets due to high brightness).
    • Synthetic dyes: Alexa FluorĀ® (488, 555, 594, 647, etc.), DyLightĀ® and tandem dyes.
